HiMedia's LP Enrichment Supplement 1 (FD214) is a component for the selective differentiation of Listeria monocytogenes from other Listeria species, particularly for use with media like HiCrome™ Listeria Ottaviani-Agosti Agar Base. It contains L-phosphatidylinositol, which is hydrolyzed by the phosphatidylinositol-specific phospholipase C (PIPLC) enzyme produced by L. monocytogenes, forming an opaque halo around colonies to visually differentiate it from other Listeria species, according to the manufacturer's Technical Data Sheet. Intended Use:
  • Selective differentiation of Listeria monocytogenes from other Listeria species. 
  • For use with media that detect PIPLC activity, such as HiCrome™ Listeria Ottaviani-Agosti Agar Base. 
Key Components:
  • L-phosphatidylinositol: The substrate that is hydrolyzed by L. monocytogenes' PIPLC enzyme. 
  • Distilled water: The solvent for the active ingredient. 
Mechanism of Differentiation: 
  • Listeria monocytogenes possesses the phosphatidylinositol-specific phospholipase C (PIPLC) enzyme.
  • This enzyme hydrolyzes the L-phosphatidylinositol in the culture medium.
  • The hydrolysis creates an opaque halo around the L. monocytogenes colonies, allowing for their visual identification.
How to Use (General Procedure): 
  • Add the sterile contents of one vial of LP Enrichment Supplement 1 (FD214) to a prepared base medium, such as an Ottaviani-Agosti Agar Base, after cooling to a suitable temperature.
  • Mix well and pour into sterile Petri plates for use in bacterial culture.
