actophenol cotton blue (LPCB) is a staining solution used in microscopy to visualize fungi. It acts as both a stain and a mounting medium, with phenol acting as a disinfectant, lactic acid preserving the fungal structures, and cotton blue staining the fungal cell walls a blue color. This allows for the easy identification of fungal elements like spores and hyphae.  Components and their functions
  • Phenol: Acts as a disinfectant, killing any living organisms in the sample. 
  • Lactic acid: Preserves the structures of the fungi, preventing them from being distorted. 
  • Cotton blue (Aniline blue): Stains the chitin in the fungal cell wall and other structures, giving them a blue color. 
  • Glycerol: Prevents the sample from drying out under the microscope. 
How it is used
  1. A sample containing fungal material is placed on a glass slide. 
  2. A drop of lactophenol cotton blue stain is added to the sample. 
  3. A coverslip is placed over the slide. 
  4. The slide is then examined under a microscope, where the fungal structures will appear blue against a paler background.
