HiMedia's Barritt Reagent B is a 40% potassium hydroxide solution used in the Voges-Proskauer (VP) test to detect the production of acetoin by bacteria. It works in conjunction with Barritt Reagent A (alpha-naphthol) to produce a red color, indicating that a bacterium has fermented glucose to produce neutral end products like acetoin. The VP test, which uses these reagents with cultures grown in MR-VP Medium, is a biochemical test for identifying members of the Enterobacteriaceae family. Function in the VP Test
  • Acetoin Detection:The VP test identifies bacteria that produce neutral fermentation end products, such as acetoin. 
  • Color Reaction:In the presence of oxygen, acetoin (from the culture) is oxidized and reacts with the alpha-naphthol and potassium hydroxide from the Barritt's reagents to form a red or cherry-red complex. 
Composition
  • Barritt Reagent B is a 40% potassium hydroxide (KOH) solution. 
  • It is part of the Barritt's reagent set, with Reagent A being the alpha-naphthol component. 
Application
  • The test is performed on 24-48 hour old bacterial cultures grown in MR-VP broth. 
  • A specific volume of Reagent B is added to the culture medium along with Reagent A. 
Interpretation
  • A positive VP test is indicated by the formation of a red color within 5-10 minutes after mixing the reagents and gently shaking to expose the medium to air. 
  • A negative test shows no red color change.
