HiMedia's Tryptophan Medium is a microbiological culture medium used for the detection of indole production by bacteria. It contains the amino acid tryptophan, which certain bacteria, like Escherichia coli and Vibrio parahaemolyticus, break down into indole. The presence of indole is then detected by adding Kovacs' reagent, which produces a red ring when positive. Different versions of Tryptophan Medium are available, including Tryptophan HiVeg™ Medium (using vegetable peptones) and Saline Tryptone / Tryptophan Medium, which adheres to specific ISO standards. Key Features and Uses
  • Indole Production Test:The primary purpose is to test for the ability of microorganisms to produce indole from the amino acid tryptophan. 
  • Bacterial Differentiation:It aids in the differentiation of bacteria, such as identifying E. coli O157:H7, a key step in coliform differentiation. 
  • Kovacs' Reagent:Indole production is identified by adding Kovacs' reagent to the culture, resulting in a red ring upon a positive reaction.

    • Tryptophan HiVeg™ Medium: A version that replaces animal peptones with vegetable-defined peptones to eliminate risks from BSE/TSE/GMOs. 
    • Saline Tryptone / Tryptophan Medium: This medium follows ISO 8914:1990 standards and is used for detecting indole production by Vibrio parahaemolyticus. 
    • DEV Tryptophan Broth: Specifically recommended for the subcultivation of coliforms and indole testing in water bacteriology, following German regulations. 
How it Works
  1. Bacteria capable of indole production are incubated in the medium. 
  2. These bacteria enzymatically break down tryptophan into indole and other products. 
  3. Kovacs' reagent is added, which reacts with the indole to form a red-colored ring at the surface of the broth. 
  4. The formation of the red ring indicates a positive test for indole production.
